The effective control of disease vectors relies heavily on chemical insecticides. Despite intense investigation of insecticide resistance in the field, several aspects of insecticide toxicology remain poorly understood. For example, the genes that influence how insecticides are absorbed, distributed, metabolized, and excreted (ADME) is imperfectly understood. Even in cases where individual genes are identified (such as insecticide metabolizing P450s), the interaction of these factors with other mechanisms of insecticide toxicology is not known. This talk will attempt to fill some of these gaps by focusing on the genetics of ADME in insect vectors with much of the findings gleaned from other insect models. We first focus on absorption of pesticide across the mosquito leg, describing our progress in understanding how the biosynthesis of the cuticle can vary between resistant and susceptible populations. Next, the distribution of insecticide around the body is described through our recent work on transporter proteins, which can increase or decrease the amount of compound arriving at its target site. Lastly, a cytochrome P450 known to act on deltamethrin is examined in the presence and absence of target site resistance, which indicates a synergistic interaction between these two alleles on the total resistance level.  While diverse on their surface, these three topics are united in their goal of understanding what factors in the insect influence the kinetics of pesticides within insect vectors.
